º» ¿¬±¸´Â Á¤Ç⠲ɺÀ¿À¸® ¿¡Åº¿Ã ÃßÃâ¹°ÀÇ ¼¼Æ÷µ¶¼º°ú Ç׿°Áõ È¿°ú¸¦ Æò°¡ÇÏ°íÀÚ ¼öÇàÇÏ¿´´Ù. ¼¼Æ÷ µ¶¼º ½ÃÇèÀº»ç¶÷ À¯·¡ ºÒ¸êÈ­µÈ Ä¡Àº¼¶À¯¸ð¼¼Æ÷ÀÎ hTERT-hNOF ¼¼Æ÷ ÁÖ¸¦ ÀÌ¿ëÇÏ¿© ¼¼Æ÷°è¼ö¹ýÀ¸·Î ½ÃÇàÇÏ¿´´Ù. Ç׿° ½ÃÇèÀºPorphyromonas gingivalis KCOM 2804 ±ÕÁÖ¿¡¼­ ÃßÃâÇÑ ÁöÁú´Ù´ç·ù(lipopolysaccharide, LPS) (PgLPS)¿Í Á¤Ç⠲ɺÀ¿À¸®¿¡Åº¿Ã ÃßÃâ¹°¸¦ hTERT-hNOF ¼¼Æ÷ ÁÖ¿¡ ó¸®ÇÏ¿©¿©, ¿°Áõ¼º »çÀÌÅäÄ«ÀÎ PGE2, IL-6 ¹× IL-8ÀÇ ¹ßÇö·® º¯È­¸¦ È¿¼Ò¸é¿ªºÐ¼®¹ýÀ¸·Î ÃøÁ¤ÇÏ¿´´Ù. ¼¼Æ÷ µ¶¼º ½ÃÇè °á°ú, Á¤Ç⠲ɺÀ¿À¸® ¿¡Åº¿Ã ÃßÃâ¹° 10~80 ¥ìg/mL ³óµµ¿¡¼­, hTERT-hNOF ¼¼Æ÷´Â82% ÀÌ»óÀÇ ¼¼Æ÷ »ýÁ¸À²À» º¸¿´´Ù. Ç׿° ½ÃÇè °á°ú, Á¤Ç⠲ɺÀ¿À¸® ¿¡Åº¿Ã ÃßÃâ¹° 10 ¥ìg/mL ÀÌ»óÀÇ ³óµµ¿¡¼­ PgLPS󸮿¡ ÀÇÇÑ PGE2 ¹ßÇöÀÌ À½¼º´ëÁ¶±º ¼öÁØÀ¸·Î °¨¼ÒµÇ¾ú´Ù. PgLPS¿Í Á¤Ç⠲ɺÀ¿À¸® ¿¡Åº¿Ã ÃßÃâ¹° 40 ¥ìg/mL¸¦ Åõ¿©Çѱº¿¡¼­ÀÇ IL-6 ¹× IL-8 ¹ßÇö·®Àº ¾ç¼º ´ëÁ¶±º(PgLPS ó¸®)¿¡ ºñÇØ °¢°¢ 75% ¹× 77% ¾ïÁ¦µÇ¾ú´Ù(p<0.05). ÀÌ·¯ÇÑ °á°ú´ÂÁ¤Ç⠲ɺÀ¿À¸® ¿¡Åº¿Ã ÃßÃâ¹°ÀÌ Ä¡ÁÖÁúȯ ¿¹¹æ ¹× Ä¡·áº¸Á¶Á¦·Î »ç¿ë °¡´ÉÇÒ °ÍÀ¸·Î »ý°¢µÈ´Ù.

This study was conducted to evaluate the cytotoxic and anti-inflammatory effects of the ethanol extract of Syzygium aromaticumL. (clove) buds. The cytotoxicity test was performed by cell counting method using hTERT-hNOF cells, a human immortalizedgingival fibroblast cell line. To test the anti-inflammatory effects, the hTERT-hNOF cells were treated with lipopolysaccharide(LPS) extracted from Porphyromonas gingivalis KCOM 2804 (PgLPS) and ethanol extract of clove buds. The expression levelsof PGE2, IL-6, and IL-8 were measured by enzyme-linked immunosorbent assay. The cytotoxicity test data showed a cell viabilityof ? 82% in hTERT-hNOF cells treated with 10 to 80 ¥ìg/mL of the ethanol extract of clove buds. The anti-inflammatory testdata showed that the expression of PGE2 by PgLPS treatment was reduced to the level of the negative control group by treatmentwith 10 ¥ìg/mL or more of the ethanol extract of clove buds. In group treated with PgLPS and 40 ¥ìg/mL of clove bud ethanolextract, the expression levels of IL-6 and IL-8 in were inhibited by 75% and 77%, respectively (p<0.05), compared to the positivecontrol (PgLPS treatment) group. These results suggest that the ethanol extract of clove buds can be used in developing oralhygine products for preventing periodontal disease.
